On Casey, Faldo said: "I was waiting for the putting to turn around. That has been very important.
"He has an extremely good record in the Ryder Cup and, going back, the Walker Cup. I feel very good about those two picks."
Clarke, well-liked by many on the PGA Tour and seen as a major part of past Ryder Cup successes, was favourite for a wildcard after winning in Holland last week.
"Nick rang me after my last round at Gleneagles to say that I wasn't going to be one of his picks and I simply wished him all the best," Clarke said.
"Obviously I am disappointed that I wasn't selected but I have dealt with much worse and it will make me all the more determined to make the team that will play in Wales next time (2010).
"I would like to wish him and his team all the very best for Valhalla. I will be supporting them from wherever I am."
Faldo also chose not to appoint a second vice-captain alongside Spain's Jose Maria Olazabal.
He had given the job to Paul McGinley, but the Irishman decided to relinquish the role in order to try - in vain - to qualify for the team.
Earlier in the day, Justin Rose, Soren Hansen and Oliver Wilson had claimed the final three automatic places in the European team after the final round at Gleneagles.
Nick Dougherty and Oliver Wilson had left themselves just too much to do to displace one of the three, but both produced rousing challenges.
Fisher shot a closing 66 and Dougherty carded a 70 in difficult conditions in central Scotland.
Montgomerie's Ryder Cup hopes hung in the balance after he failed to make an impact on the last day of the Johnnie Walker Championship.
The Scot ended on one over at Gleneagles.
"If I get selected it'll be fantastic. If I don't I wish the team every success," said the 45-year-old.
Europe have won the last three matches and five of the last six in the biennial competition with the US.
The 37th Ryder Cup takes place at Valhalla Golf Club in Louisville, Kentucky, from September 19-21.
Four of the 10 automatic qualifiers - Rose, Wilson, Hansen and Northern Ireland's Graeme McDowell - will be making their Ryder Cup debuts at Valhalla.
